JMJD1C encodes a jumonji-domain-containing histone demethylase that removes methyl marks from histone H3 lysine 9 (H3K9me1/2), acting as a transcriptional coactivator for nuclear hormone receptors. Common variants at the JMJD1C locus, such as rs7910927, are among the strongest known determinants of circulating sex hormone-binding globulin, the carrier protein that transports testosterone and estradiol in the blood. Because SHBG binds testosterone, these variants also shift measured total testosterone — but large studies find no effect on the free, biologically active fraction in men.
