rs115366859 is an intronic variant in GC on chromosome 4, the gene encoding vitamin D-binding protein (DBP), the primary plasma carrier of 25-hydroxyvitamin D. In a gene-by-environment GWAS of over 338,000 UK Biobank participants (Shraim et al., Nat Commun 2025;16:10774), it had the largest main effect on 25-hydroxyvitamin D among newly identified variants (beta -0.154, p 5.4e-50). It does NOT interact with sun exposure: in the same study's deposited interaction statistics its gene-by-UVB p-value is 0.72. The variants that did show a UVB interaction sit at CYP2R1, PDE3B, PSMA1, COPB1 and CALCB, 80% of them clustered on chromosome 11 — not at GC.
